The 5km run or walk in Horsham, which attracts about 300 participants, is in its 15th year.
Since 1998, the tally from all Mother's Day Classics across the country has topped $44 million.
The motto for the 2024 walk or run is: Move 'til we’re done.
Organiser Jackie Exell is urging runners and walkers to become involved to help support these causes.
The event starts on Mother's Day, Sunday May 12, at 9am, and starts and finishes at Horsham Soundshell.
Ms Exell said she first joined the event as a "get healthy" kick with a friend.
She has been inspired by her grandmother, Hazel Exell, who had a single mastectomy in 1975 after a breast-cancer diagnosis, and is still fit and well at 99. She lives at Kurrajong Lodge, Horsham.
"Women die each day from cancer," Ms Exell said. "And we're excited that the Mother’s Day Classic Foundation is including ovarian cancer research for the first time."
Ovarian cancer is often undetected until it has spread within the pelvis and stomach, when it is more difficult to treat and can prove fatal.
Ms Exell urged people to start their Mother’s Day "with purpose" and join the nearest classic event.
She is also encouraging businesses to register runners - there is a prize for the team with most participants.
Everyone who registers online before Friday May 10 will go into a draw for a necklace from Mackays Family Jewellers.
"And on the day a barbecue breakfast will be available, a coffee van will be on hand, there is face painting for children and spot prizes will be given out," Ms Exell said.
